Swapping restriction enzymes

You can quickly swap restriction enzymes in the Breeze BioCAD by either replacing the bases in the sequence editor or swapping biomodules.

The Modular Way


1. Find placeholder for RE sites.

To swap restriction enzymes, "the modular way", you must ensure you are designing with a modular construct that contains placeholders for Restriction Enzymes or Hinge sites.

Select the construct you would like to edit.

Choose to view the construct in the basic view.

Confirm the restriction enzyme site is contained within a placeholder by hovering over that placeholder.

2. Remove the restriciton enzyme biomodule

Once you have identified the placholder for your Restriction Enzyme site, you can remove the biomodule in that location in order to swap it.

Click on the placeholder containing your RE biomodule.

Click remove.

3. Replace the Restriction Enzyme biomodule

Now you have removed the previous biomodule, you can replace it with another biomodule containing restriction enzyme.

Once, you have removed the previous biomodule, you will be presented with your biomodule collection. This collection is automatically organised by relevance according to the functionality of the placeholder you are editing. In this instance, biomodules containing Restriction Enzymes will sorted first.

Use the search feature to search the name of DNA sequence of the Restriction enzyme biomodule you would like to add.

Via the Advanced View


If the design you are working on does not contain individual biomodules for your restriction enzymes, you can still edit them, using the replace bases tool in the advanced view.

Navigate to your restriction enzyme location

In the advanced view of the construct designer (or standard view of the biomodule editor) use the annotation navigation sidebar to find the Restriction Enzyme site you would like to swap.

Jump to that location by clicking on the arrow for that RE site.

Ensure you have selected the entirety of the restriction enzyme site. (Click and drag to make a selection)

Right click on the DNA sequence and choose "replace bases".

In the sequence module, type the sequence of the restriction enzyme you would like to replace it with.
